Welcome to the site! Treadmill will help you lose fat to make you abs and glutes more defined but won't make your glutes any bigger.

The best way to train the glutes imo is to do exercises that train the entire posterior chain.

squats including things like lunges
deadlifts - reg, romanian
pullthroughs
glute-ham raises
reverse hyperextensions
